The ability to grow human tissues from stem cells in 3D culture has the potential to revolutionize the drug discovery process and regenerative medicine. Building on a long tradition of cell and developmental biology knowledge, organoids resembling a variety of human tissues have been generated. This symposium will bring together the leading researchers in this field to establish a new research community and reveal parallels between various tissue models.

The aim of this meeting is to bring together researchers from different fields to enhance our understanding of how organoids can be formed and maintained, how they can be used to study disease and how we might eventually use them to regenerate and replace human organ tissue.

Organ-on-a-chip with organoids: the best of both worlds

The transition from 2D to 3D cell culture is a first step towards more physiologically relevant in vitro models for human disease modeling. Organ-on-a-chip and organoid technology are two fields of development that are rapidly redefining cell culture biology in the 21st century.

Organ-on-a-chip technology has demonstrated how microengineering can add aspects such as perfusion flow, patterning of tissues, extracellular matrix embedment, vascular structure, precise control over gradients and application of mechanical strain to tissue culture. Organoid protocols have been carefully optimized to maintain the stem cell niche in developing and adult tissues, while at the same time permitting cell proliferation and differentiation. MIMETAS develops organ-on-a-chip-based models for disease modeling and evaluation of new medicines. The OrganoPlate®, a unique microfluidic technology, enables testing of compounds on miniaturized 3D organ models in high-throughput.

Combining this complex biology with advanced high-content imaging techniques, such as those enabled by the ImageXpress Micro Confocal System (Molecular Devices), opens up a whole new level of assays. A powerful automated confocal imaging instrument equipped with advanced 3D analysis capabilities can allow researchers to obtain accurate, quantitative results, and answers to their questions, very quickly and in a robust and scalable manner.

In this webinar, we will present some of the latest developments in integrating organoid protocols with organ-on-a-chip technology, as well as advancements in the high-content imaging technologies that are enabling these advanced 3D applications. We’ll show how a range of tissue models comprising complex co-cultures can be formed in a perfused system using state-of-the-art stem cell and organoid protocols, and how such models can then be screened and analyzed within a single, integrated interface to dramatically reduce time to discovery.
